Q: Will 2 1/4 x 4 cards work?

Not unless the 4" side is a bit less than 4" wide. Inside diameter is jusr over 3 7/8" The 2 14 measurement works.

Q: Does it come with the dividers?

No it did not come with the dividers. The box was larger than I expected & will hold a lot of cards. It was too big for what I needed & I could not use it. I liked the purple color, but nothing else.
