Is this package enough to balance 2 Light Truck tires?

There are 72 one once weights in a box. It generally takes about 6-8 ounces of weight to balance a cheap tire on a not-so-good wheel so you will have way more than you need for two tires.

Can you put the weights on leather and then we move them at a later date

I would not recommend to do so. These will be hard to remove from leather.

Can these weights withstand up to 275-300 F?

The metal parts are fine for this temperature; however the service temperature of the tape is best in the range of -20 to 150F.

Will these work for aluminum semi truck wheels as well??

Yes, they would, except that semi truck tires can need a lot of weights.
