Does this case fold out of the way for holding the tablet comfortably in portrait mode?

The only way to use the case to view the tablet in portrait mode is to prop it up on your knees and hold with your hands. In other words, it does not swivel to portrait mode you have to hold it that way yourself and stabilize it yourself.

Will the S pen fit in the slot with a case on it?

There is a plastic slot in the case, with retention for the pen. It also protects the pen from falling out when the case is closed.
