Can this be used in refrigerator and freezer?

I'm not sure about a freezer but most likely could be used in a refrigerator. I have one in my garage, it's in there all summer and winter. I live in NY, so it's either really hot or cold.

How do you calibrate humidity?

To the best of my knowledge, they cannot be calibrated. I've placed several in the sale location and have gotten only a few percentage points variance between the hygrometers.

Will this give me temperature inside empty ice chest

Yes it will. Place the instrument on a piece of plastic about an inch high. By not having it on the bottom it will give you the air temp and rh inside the chest.
