Can these be sealed and resealed again and again or are only good for sealing an envelope for only one time?

I wouldn't recommend it. You can remove and try and reseal, but they don't stick as well the second time, I tried it and had to put another sticker over it.

They are a little difficult to remove from the roll, any suggestions?

Yes, they are difficult to remove from the roll. I don’t have any suggestions other than using your thumb nail.
