No, these do not have a microphone.

How do I tighten this earphone after I clip on?

You don't. It only has two positions, open and closed. You can raise and lower the ear-clip which might help it to rest more securely against your ear, but it should rest comfortably against the side of your head without falling off in most circumstances. It might not be appropriate for running, but I used to wear the aluminum variants when I worked out at the gym without them falling off, for example.
