are the pieces plastic or wood or what?

Some are plastic and the disks are sturdy cardboard.

Hello! My daughter lost one potion bottle. How can I get replacement pieces for this game?

Peaceable Kingdom will send you lost pieces free of charge as long as they're still making them. Request it through their customer service.

Would this game be too simple for kids turning 9 and 12?

my 8 year old likes it so i think your 9 year old would still enjoy it... i wonder if it might be simple for the 12 year old though?

Is this game good for four year olds?

This a great beginner game for young kids 4-8 because there are no real winners or losers per se. 4 would be about the youngest I’d think would enjoy it. It’s very cute and simple, and as I remember, I believe players work collaboratively, which avoids the hurt potential hurt feelings that result from more competitive games.
