Can this handle 600 volts typically found in restoring tube radios. I would think it would be the same as for tube amps.

This wire is specifically manufactured for use in guitars, and thus, the manufacturer (Gavitt) does not publish an insulation resistance rating. It is also stranded and most vintage gear uses solid core wire, typically 20 gauge or larger (this is 22awg, which is smaller). Many suppliers of vintage "style" amplifier repair wire suggest and sell cloth covered PVC insulated wire for repairs and new builds. This wire has only a waxed cotton insulator, so I would not recommend using it in a high voltage application as there are far safer and similar "looking" products available that have modern insulation properties.

Does this wire require any special shielding for use in a Les Paul guitar?

Although this type of wire is traditionally used in Fender guitars, it can be used in any guitar, but it is not shielded like the wire that is typically used in a Les Paul. 
Most Les Paul guitars would use Gavitt Vintage Style Braided Shield Hook Up Wire For Guitars, which has a single conductor pushback wire with an outer braided shield that is used for the ground side of the circuit. You may want to Google some photos of Les Paul Control Cavities.
Modern Les Paul guitars  use a circuit board that the pickups hook directly in to and may not use outside braided wire at all (depending on which pickups are used). 
My opinion is "the more shielding, the quieter the guitar will be". To wire a Les Paul in the "traditional" configuration, you would use outside braided wire instead of unshielded wire, but I have seen many Les Pauls that have used "other" wire.

I just checked what I have left over after a pickup wiring project. I have about 5.5 feet of each color so it was definitely 6 ft of each. The ad's 3 foot statement is a typo.
